چکیده

In a previous publication on the Gram staining material of human epidermis we advanced the theory that keratohyalin contained a highly resistant mucopolysaccharide. In the course of normal keratinization this material was decomposed, while in parakeratotic and many other types of pathologic scales it was retained in the horny layer (1). The evidence presented was indirect: we found that the extent of Grampositivity roughly paralleled the hexosaminet content of hydrolyzed horny layers and mercuric salts which blocked the Gram stain, also greatly reduced the amounts of reactive hexosamine in hydrolyzed scales. We were unable to provide more direct evidence, because this highly resistant substance defied attempts at isolation or selective destruction. In our recent experiments we were able to overcome some of these difficulties. Partial isolation, purification and enzymatic destruction of epidermal mucopolysaccharides will he briefly discussed in this preliminary report.

Study of epidermis development in sturgeon (Acipenser persicus) larvae.

Fish skin is essential for survival, maintenance of body shape, and protection against the shock and infection. During week 1 of sturgeon larval development, the epidermis is thin and not differentiated in various layers yet, but by week 4-6 the thickness increases and various layers appear, depending upon the region of the body. Mucous cells differentiate early in development on the surface of...

Biochemical Methods in Production of Three-Dimensional Scaffolds from Human Skin: A Window in Aesthetic Surgery

BACKGROUND Use of matrix-derived biologic scaffolds has become a treatment of choice in several clinical issues. This study assessed biochemical methods in production of three-dimensional scaffolds from human skin. METHODS Human skin was prepared from circumcisions, washed in phosphate buffer saline (PBS) and kept at -20ºC until use. The skin samples underwent various methods. In group A, Na...

Mucopolysaccharides Produced in Tissue Culture

1. A method of mass tissue culture has been devised by which, in a relatively short period of time, samples large enough for chemical isolation of mucopolysaccharides can be obtained. 2. Chemical isolation of acid mucopolysaccharides from mass cultures of human fetal skin, human fetal bone, bovine fetal skin, and rat subcutaneous tissue has been carried out. It has been found that the fibroblas...

Histochemical Studies of Human Coronary Atherogenesis: Comparison with Aortic and Cerebral Atherogenesis.

• The literature contains many conflicting reports concerning the role of acid niucopolysaccharides and lipids in the pathogeneyis of coronary atherosclerosis. The most notable controversy concerns the earliest morphological alteration in atherogenesis. Some investigators hold that the accumulation of acid mucopolysaccharides is the reaction to a primary deposition of lipid, while others hold t...

Blocking of Hog Intrinsic Factor by Human Gastric Juice and Certain Mucopolysaccharides , in - Cluding Blood

Intrinsic factor can bind vitamin B12 and simultaneously attach to receptors on liver or small intestine in a calcium-dependent manner (1). There appears to be a stoichiometric relationship between the receptors, intrinsic factor and vitamin B12 (2). This report, previously presented in preliminary form (3), is concerned with blocking of the receptor sites for hog intrinsic factor by human gast...
